The Technology

Small RNAs are a 20-30b long RNA molecules which play an important role in regulation of gene expression. One of the most important type of small RNAs are microRNAs. MicroRNAs are conserved and function in RNA silencing and post-transcriptional regulation of gene expression. One of the common methods to identify and quatnify small RNA in cells is to sequence the small RNAs by using next-generation sequencing platforms. For this purpose ” small RNA Library” is prepared by extracting small RNAs from cells followed by its ligating to “linker” molecules from both sides. The obtained library is then amplified by PCR and subjected to High-throughput sequencing (HTS). The sequence data is then analyzed to obtain the abundance and location of each small RNA in the samples. The invention is a new method for of small RNA library preparation and analysis process of the sequence data.
